有关脂肪酸甲酯化的问题?最近在做细菌脂肪酸的提取和测定,需要将 脂肪酸甲酯 化后跑GC-MS,查阅了相关的文献,发现用的最多的是Morrison and Smith(1964)的方法,即 三氟化硼甲醇 溶液甲酯化,文献中有这么一段话Boron fluoride-methanol reagent was added under nitrogen, in the proportions 1 ml reagent per 4-16 mg of lipid, and the tube was closed with the screw cap. The tube was then heated in a boiling water bath for the requisite time, cooled, and opened. The esters were extracted by adding 2 volumes of pentane, then 1 volume of water, shaking briefly, and centrifuging until both layers were clear.三氟化硼不是和水剧烈反应么,这里加水会不会和三氟化硼反应啊还有就是这篇文献就是将三氟化硼甲醇溶液加到脂肪酸里拧上盖子100摄氏度水浴2min就可以,这和国标用冷凝管回流什么的简化了不少,能达到良好的甲酯化效果么,我看到有的文献还有在瓶子里拧上盖子100度水浴40min的 = = 第一次做相关的实验完全没经验啊,大神们求解答啊,谢谢谢谢查看更多7个回答 . 10人已关注
