C. 6 g of iodide was added to the reaction mixture in small portions over a period of 2 hours. The reaction mixture was stirred and heated for 7 hours. The reaction mixture was cooled down and precipitated. Crystals were filtered off on a Buchner funnel and washed with water. The product was crystallized from ethanol (50 mL) to yield 5.57 g of the desired material. <BR><BR>5-(1-methylbutyl)-5-iodoacetonyl Barbituric Acid <BR><BR>1 g of 5-(1-methylbutyl)-5-(.beta.-hydroxy-v-iodopropyl) barbituric acid was dissolved in 75 mL of acetone. 30 mL of a 0.33 mol solution of potassium dichromate in 10% sulfuric acid was added. The reaction mixture was stirred for 2 hours at room temperature and subsequently extracted with 200 mL of ethyl acetate, washed with brine, water and the organic layer was dried over magnesium sulfate. Ethyl acetate was removed in vacuo and crude solid material crystallized from 70% ethanol. The yield was 519 mg of colorless crystals of the desired product. <BR><BR>Coupling of 5-(1-methyl)iodoacetonyl Barbituric Acid to Bovine Serum Albumin <BR><BR>226 mg of BSA was dissolved in 4.7 mL of phosphate (buffer 0.1 M, pH 8) and 520 ul of N,N'-dimethylformamide. To this solution was added 152 mg of 5-(1-methylbutyl)-5-iodoacetonyl barbituric acid in 530 uL of DMF. The reaction mixture was stirred at room temperature for 36 hours.
